Preparation

# Dissolve 58g in 1000ml of distilled water (purified water)

# Heat and frequent stirring to completely dissolve the medium

# Autoclave at 121°C for 15 minutes

# Mix well and dispense into appropriate sterile containers

Description

ChromoGel™ E coli Coliforms Agar is a selective medium used for the isolation and identification of E.coli and Coliforms from clinical specimens, foodstuff and environmental samples.

  • The medium can use as secondary selective agar plate on ISO method, petri-film method.
  • The samples can streak directly on surface agar plates without enrichment step.
  • Aerobic Incubate:

– At 30 ± 2°C for 18-24 hours for Total Coliforms

– At 37 ± 2°C for 18-24 hours for E.coli and Coliforms

– At 44 ± 2°C for 18-24 hours for faecal Coliforms

  • Read the result within 18-24h

 Technique

Membrane filtration method:

  1. Filter the sample on a 0.45μm membrane and put into surface agar plate.
  2. Aerobic Incubate: 18-24h
  3. Count as E. coli and coliform confirmed by indole test.

 Streaking method:

  1. Streaking 1-10µl of the sample on surface agar plate.
  2. Aerobic Incubate: 18-24h
  3. Count as E. coli and coliform confirmed by indole test.

 Spreading method:

  1. Spreading 0.1-1ml of the sample on surface agar plate.
  2. Aerobic Incubate: 18-24h
  3. Count as E. coli and coliform confirmed by indole test.

Chuẩn bị

# Hòa tan 58g trong 1000ml nước cất (hoặc nước đã lọc)

# Gia nhiệt và khuấy thường xuyên để môi trường hòa tan hoàn toàn

# Khử trùng ở 121° C trong 15 phút

# Lắc đều và rót vào các dụng cụ chứa vô trùng thích hợp

Đặc tính sản phẩm

ChromoGel™ E.coli Coliforms Agar là môi trường chọn lọc dùng trong nuôi cấy định danh e.coli và coliforms từ mẫu bệnh phẩm, thực phẩm và mẫu môi trường.

  • Môi trường được dùng như đĩa thạch đối chứng thứ 2 dựa theo phương pháp ISO hoặc Petri-film
  • Mẫu kiểm có thể được cấy trực tiếp lên bề mặt đĩa thạch không cần qua tăng sinh.
  • Nuôi cấy hiếu khí:

– Ở 30 ± 2°C for 18-24 giờ cho tổng Coliforms

– Ở 37 ± 2°C for 18-24 giờ cho E.coli và Coliforms

– Ở 44 ± 2°C for 18-24 giờ cho  faecal Coliforms

  • Đọc kết quả trong vòng 18-24h

  3. Đếm khuẩn E.coli và Coliforms và định danh bằng Indol test

Phương pháp cấy ria:

  1. Ria 0.1-1ml mẫu trên mặt thạch
  2. Ủ hiếu khí 18-24h
  3. Đếm khuẩn E.coli và Coliforms và định danh bằng Indol test
