Preparation

# Dissolve 65g in 1000ml of distilled water (purified water)

# Heat and frequent stirring to completely dissolve the medium

# Heat with frequent stirring until boiling (DO NOT AUTOCLAVE)

# Cool to 45-50°C, mix well and dispense into appropriate sterile containers

Description

The medium can be used according to usual laboratory techniques with incubation at 35°C-37°C and results reading after 24 and 48 hours. The chromatic characteristics of the colonies after 48 h of incubation at 35°C-37°C are the following:

• green, blue-green colonies: characteristic of C.albicans
• blue-grey colonies with violet shade and/or violet halo: characteristic of C.tropicalis
• pink, pink-violet, large, rough, spreading, colonies: characteristic of C.krusei
• White or pink or pink-violet colonies: characteristic of other Candida species.

Confirm and complete the colonies identification with suitable tests.

 Technique

Membrane filtration method:

1. Filter the sample on a 0.45μm membrane and put into surface agar plate.
2. Aerobic Incubate: 18-24h
3. Count Candida bacteria

Streaking method:

1. Streaking 0.1-1ml of the sample on surface agar plate.
2. Aerobic Incubate: 18-24h
3. Count Candida bacteria

 Spreading method:

1. Spreading 0.1-1ml of the sample on surface agar plate.
2. Aerobic Incubate: 18-24h
3. Count Candida bacteria

Storage

Store the medium tightly closed in a dry place at room temperature and use before the expiry date on the label.

Chuẩn bị

# Hòa tan 65g trong 1000ml nước cất (hoặc nước đã lọc)

# Gia nhiệt và khuấy thường xuyên để môi trường hòa tan hoàn toàn

# Đun nóng và khuấy thường xuyên cho đến khi sôi (KHÔNG HẤP VÔ TRÙNG)

# Làm mát đến 45-50° C, lắc đều và rót vào các dụng cụ chứa vô trùng thích hợp

Tính chất môi trường

Môi trường được sử dụng nuôi cấy định danh vi nấm Candida được ủ ở 35°C-37°C  trong 24-48 giờ. Enzyms chromogenic tạo màu vi nấm sau thời gian ủ sẽ cho kết quả như sau:

• Candiada albicans sẽ có màu xanh lá green
• Candida tropicalis sẽ có màu xanh dương với vòng xung quanh màu tím
• Candida krusei sẽ có màu tím đậm, khuẩn lạc lớn.
• Các dòng candida khác có màu trắng hồng hoặc hồng
